Linguistic human rights are not strictly essential human rights; they exist to improve on the basic human and civil basic right to language. Optimizing the condition of the PCR detecting of PTEN gene in DNA derived from human cell lines, associated with ageing and senescence - Research Paper Example Traditional PCR is used for amplifying DNA for sequencing, genotyping and cloning. However, this has a number of shortcomings namely; poor precision, low sensitivity, short, dynamic range of less than 2 logs, low resolution, is non-automated, based on discriminatory size only, results cannot be expressed as numbers, the staining agent ethdium bromide is not efficient at quantification and finally, post-PCR processing is required. This paper looks at the analysis of PTEN using traditional PCR method. The process was not successful despite the necessary steps being undertaken. PTEN primer, when used on a Hacat cell line, showed annealing at approximately 62 degrees. However, the use of PTEN with similar parameters that were later altered for optimization yielded no results. The methodology has been discussed, and the gel results showing synthesized images have been included in this report. The discussion that covers the PCR process and its effectiveness has also been included. The fidelity of the PCR process has been discussed to extrapolate on the possible reasons for the failure of this analysis. Phosphatase and Tensin Homolog gene commonly referred to as PTEN gene provides the necessary instructions for the synthesis of a protein that is present in almost all body tissues (Sulis, 2003). The protein is significant in its function as a tumor suppressor and aids in the regulation of cell division by inhibiting cells from diving and growing too fast or uncontrollably. PTEN is a phosphotase which modifies other fats and proteins through the removal of phosphate groups comprising of one phosphorus atom and three oxygen atoms (Cohen 1990). Negligence of Tort Essay DEFINITIONS: In common law jurisdictions, tort is a civil wrong although in certain circumstances it is also treated as a criminal case e.g. in cases of assault. Duties imposed under tort law are considered mandatory for all citizens unlike voluntary obligations imposed under contracts. If a person commits a tortuous act, he/ she are legally a Tortfeasor. Tortiuos acts include breach of duties imposed under statutory laws, causing bodily harm to other people, property, or infringing on another’s legal rights.for there to be a case under tort NEGLIGENCE: This is a legal concept that is usually used to acquire compensation for injuries suffered or accidents met. It is a civil wrong actionable under tort law. Negligence involves behaving in a manner that lacks the legality of protecting other people against foreseeable risks. Under common law for there to be a case under negligence then the following elements must be satisfied. * Duty of care * Breach of the duty EXAMPLES: Experience Of Harm Arising From That Breach. Donoghue V. Stevenson A case of tort on negligence is only successful if the following five elements are met. Firstly, a duty of care must be evident. Donoghue v. Stevenson. This is because the company’s negligent actions are the reason why each involved party is at a loss. The company should prepare itself for lawsuit and the family friendly fishing company should brace itself, to be legally prepared to pay special damages, general damages as well as punitive damages. On top of this, the company should also be prepared for the courts to issue an injunction against them if it is deemed necessary. CONCLUSION: The five elements of negligence should be recognizable under tort law. However, as professor Robertson of Texas University has noted the five elements needs to be kept separate, this is because in practice there arises lots of conceptual mistakes or confusions. The use of vicarious liability as a defense against a case of law on tort of negligence has widely been disputed over its rationale. Many scholars have argued that the use of vicarious liability is only intended to find legally  a more solvent defendant. In a legal perspective, the family friendly fishing company is supposed to pay special and general damages to Gina and June. This is because the company has caused the two irreparable damage because of its own negligent activities. Damages awarded are usually compensatory but not punitive in nature. For the sake of the family friendly fishing company, the damages it would have to pay would most probably include a punitive element. The punitive element would arise if the employee, Neil would declare that indeed he had asked the company to change the boats gear, although the company declined. This refusal of the company to use standard tools for profiteering reasons would be termed as an act of maliciousness or acting in an callously indifferent way.

Both of the activities directs towards achieving the appropriate candidate with the requisite competencies and attitudes. The recruitment can also be understood as the foundation for the selection process by providing the number of applicants from which the selection panel and select the suitable candidates. So it is the source of manpower to meet the requirements which are needed in the staffing (Flippo, 2000). There are basically two types of recruitment, internal and external. The countrys economic policy framework reflects features such as structure, size and diversity of the banking and the financial sector. There are basically two types of sectors operated in India, Private sector and the Public sector. These companies are well known in national and as well as the international companies. The people are the main force on which the economy depends. If the HR people take a wrong decision in selecting a candidate to work the company can die. The reason of retention in the companies, as the research requires more of interviews, which can be telephonic or face to face makes the report more descriptive. "Discovering Communities on the Internet" Section I Online Communities Online communities seem to be very popular these days. I feel there is two different types of online communities. One type is where people converse during real time, meaning an individual could typically ask a question and will receive an immediate response from someone else. The immediate responses result in a dialogue between two or more people. Bulletin Boards are also popular because they are similar to an online community. The bulletin boards allow people to share ideas. To my understanding, the main difference between the two is that someone posts an idea on the board, and another person responds to the inquiry or statement by posting his response. Both of these places allow people to have discussions of all kinds and for a variety of reasons. Some people may think that an online community is where they met their significant others. Other people think an online community allows them to find the house or car of his dream. Also, people will ask for advice or ideas about something that is going on in life. There are advantages and disadvantages in chatting with communities. Some of the advantages are as follows: people are connected with the world and the diversity of people; people are able to find out other people's interests and ideas; people can learn about a different person's perspective. Some disadvantages in chatting with communities are as follows: people can learn how scary and different people really are; people can lie to you with out you even knowing the truth. I have had personal experience with chat communities and I have found great success. I have used chat communities for educational purposes. When I have chatted with communities I felt at ease. For example instead of having a class discussion I chatted with my classmates online. This way of discussing ideas was neat, because at first I did not know who I was talking to, then eventually I did figure out who I was chatting with. Another reason why I have enjoyed chat room discussions versus in class discussions is because people had the opportunity to speak their minds over the computer and they were not hesitant or shy. People did not have to face humiliation from the teacher or other classmates by their vocal answer. Chat rooms allow everyone to be able to participate and get involved in the class.

The Joy Luck Club 1. From the film The Joy Luck Club, Chinese girls were supposed to act obedient and respectful to their parents and elders. This included the girls having to abide by each and every Chinese tradition that their parents instilled in them. Girls were also expected to be quiet and considerate to their parents and elders. They were only supposed to speak when spoken to at all times. Acting out against anything their parents enforced upon them was completely unacceptable. 2. Much like to their parents and elders, Chinese girls were supposed to be quiet and obedient to their husbands and mothers-in-law. Girls were supposed to respect, and not openly question the decisions that were made in the household. These women were taught to always ask what the husband wants and to ignore their own wants and needs. Chinese wives were there mainly to provide the mother-in-law with grandsons. 3. Chinese victims of rape were treated with complete disrespect and disbelief. As with the one mother in The Joy Luck Club, she was kicked away by her own family for making such a claim. These rape victims were seen as disrespectful to their families and themselves. One social consequence of claiming rape is that their families shun the women and force them to leave their homes. Secondly, the community shuns them as well. As with the woman in the film, she was denied work and abandoned by her neighbors. She was eventually forced to marry her assailant in order to save her child. 4. One of the main psychological consequences of having the system of many wives and concubines is that not only the husband, but also wives have complete power over each other as in a hierarchical system. As in the film, the secon... ... did not afford her these things, Lindo is being very cautious, often critical, of her daughter and the choices that she has. 10. The daughters in this film struggled with traditional sex roles mainly due to the influence of their mothers. In many cases, the mothers tried desperately hard to encourage their daughters to have power over their lives, be successful, and have a strong self-esteem. This over-encouragement to lead a life that they could not, led many of the daughters to feel inferior because they could not live up to their mother's expectations. In some cases, this led to the daughters getting involved in relationships in which they relied on their husbands for power and support. So in essence, it was their mothers' extremely high hopes for their daughters that led them to feeling inferior as women when these hopes could not be fulfilled.
